We are currently trying to estimate the population sizes of three populations of the rock firefinch Lagonosticta sanguinodorsalis as a part of a conservation genetic study within Jacey Abalaka’s PhD project. We were very pleased to see the updated version of NeEstimator presented in Molecular Ecology (Do et al. 2014, doi: 10.1111/1755-0998.12157). Both single- and two-sample estimators are available, and we are using the latter method since we have data from different time periods and also since we believe that this method is the superior one. It is still not absolutely clear whether to estimate F as suggested by Nei & Tajima, Pollak or Jorde & Ryman – but judging from the variation in the parameter estimates, we chose Pollak’s method. Not surprisingly, we get quite low Ne:s with large confidence intervals in our populations.
